    Chloroquine nuclei acid biosynthesis

Indeed, the ability to salvage nucleotides from sources within the body alleviates any significant nutritional requirement for nucleotides, thus the purine and pyrimidine bases are not required in the diet. The salvage pathways are a major source of nucleotides for synthesis of DNA, RNA and enzyme co-factors.

Chloroquine Resochin was synthesized by Andersag in 1934 rev. by Coatney, 1963 and became the prototype of the 7-halogenated 4-aminoquinoline antimalarials which, as a group, were patented by Andersag et al 1939. The chemical structure of chloroquine is shown in Fig. 1. Chloroquine or hydroxychloroquine can also directly bind to nucleic acids and hence might block TLR9 signalling at the intracellular level by inhibiting TLR–ligand interactions steric blockade. NNuucclleeiicc AAcciiddss Nucleic acids are molecules that store information for cellular growth and reproduction There are two types of nucleic acids - deoxyribonucleic acid DNA and ribonucleic acid RNA These are polymers consisting of long chains of monomers called nucleotides A nucleotide consists of a nitrogenous base, pentose sugar and a phosphate group.

Following their synthesis in the liver the nucleotides are dephosphorylated and in part phosphorolytically cleaved into nucleobases and ribose-1-phosphate for transport to the blood and then subsequent uptake by cells of the other organs. Within the body the major site of de novo nucleotide synthesis, for the replenishment and maintenace of intracellular pools, is the liver.

Chloroquine is a 4-aminoquinoline with antimalarial, anti-inflammatory, and potential chemosensitization and radiosensitization activities. Although the mechanism is not well understood, chloroquine is shown to inhibit the parasitic enzyme heme polymerase that converts the toxic heme into non-toxic hemazoin. The nucleic acids of two resistant strains of Plasmodium falciparum were studied by acridine orange staining and differential fluorescence of DNA and RNA. Treatment of the Thailand strain with pyrimethamine, to which it is resistant, produced no reduction of RNA, and only slight reduction of DNA. IMP, whose de novo biosynthesis we have just seen, is not a normal constituent of nucleic acids; it will be converted into adenylic and guanylic nucleotides, which are the major purine nucleotides found in ribonucleic and deoxyribonucleic acids. These transformations are diagrammatically depicted in figure 6-21.
